Dietary Geranylgeranyl Pyrophosphate Counteracts the Benefits of Statin Therapy in Experimental Pulmonary Hypertension

چکیده

Background: The mevalonate pathway generates endogenous cholesterol and intermediates including geranylgeranyl pyrophosphate (GGPP). By reducing GGPP production, statins exert pleiotropic or cholesterol-independent effects. potential regulation of homeostasis through dietary intake the interaction with concomitant statin therapy is unknown. Methods: We developed a sensitive high-pressure liquid chromatography technique to quantify conducted proteomics, qualitative real-time polymerase chain reaction screening, Western blot determine signaling cascades, gene expression, protein–protein interaction, protein membrane trafficking in wild-type transgenic rats. Results: contents were highly variable depending on food source that differentially regulated blood levels Diets containing intermediate high reduced abolished effects rats hypoxia- monocrotaline-induced pulmonary hypertension: this was rescuable by methyl-allylthiosulfinate methyl-allylthiosulfinate–rich garlic extracts. In human artery smooth muscle cells treated statins, hypoxia activated RhoA an extracellular GGPP-dependent manner. Hypoxia-induced ROCK2 (Rho associated coiled-coil kinase 2)/Rab10 (Ras-related rab-10) prevented recovered exogenous GGPP. hypoxia-activated RhoA/ROCK2 rat upregulated expression Ca 2+ -sensing receptor (CaSR) HIMF (hypoxia-induced mitogenic factor), mechanism attenuated treatment regained Rab10 knockdown almost abrogated hypoxia-promoted CaSR trafficking, process diminished resumed hypertension mutated at binding motif between efficiency abolished. humans fed diet, increased. This statin-lowering plasma GGPP, also hypoxia-enhanced activity monocytes rescued Conclusions: There important interferes experimental hypertension. These observations rely key central role RhoA-ROCK2 cascade activation Rab10-faciliated subsequent overexpression CaSR. findings warrant clinical investigation for perhaps other diseases combining garlic-derived extracts thus circumventing variations.

Benefits from the correction of vitamin D deficiency in patients with pulmonary hypertension

Background: Vitamin D (Vit D) is linked to various conditions including musculoskeletal, metabolic and   cardiopulmonary diseases. However, it is not clear whether correction of vit D deficiency exerts any beneficial effect in patients with pulmonary hypertension. Methods: This study was a prospective uncontrolled longitudinal study. Patients with pulmonary hypertension and vit D d...

Statin-induced depletion of geranylgeranyl pyrophosphate inhibits cell proliferation by a novel pathway of Skp2 degradation

Statins, such as lovastatin, can induce a cell cycle arrest in the G1 phase. This robust antiproliferative activity remains intact in many cancer cells that are deficient in cell cycle checkpoints and leads to an increased expression of CDK inhibitor proteins p27Kip1 and p21Cip1. The molecular details of this statin-induced growth arrest remains unclear. Here we present evidence that lovastatin...

Drug Therapy in Patients with Chronic Thromboembolic Pulmonary Hypertension

Background: Chronic Thromboembolic Pulmonary Hypertension (CTEPH) results from the obstruction of predominantly major pulmonary vessels by organized blood clots and is one of the causes of pulmonary hypertension. The disease is still underdiagnosed and the true prevalence is not clear. The objective of this study was to determine the clinical characteristics and current management of patients w...

Dietary pulmonary hypertension.

In 1967 Kay et al postulated that some cases of unexplained pulmonary hypertension might be due to the ingestion of drugs or other toxic substances.' This concept, later termed dietary pulmonary hypertension,23 was advanced following the observation that the oral administration to rats of the pyrrolizidine alkaloid monocrotaline produced severe pulmonary hypertension.' There are no reports of p...
